J. Raymond DePaulo, Jr., M.D. (Distinguished Investigator 2003) of Johns Hopkins University, has studied families with bipolar disorder (BP) for the last 15 years, and now proposes to conduct genome scans on subjects available to him through these samples. From one of his previous studies, he has selected the region with the greatest evidence of a linkage to BP, and will use Single Nucleotide Polymorphisms (SNP) genotyping to capture the linkage disequilibrium information in a region identified by Dr. DePaulo and other researchers (locus at 8q24). He will use the SNP genotypes and the haplotypes derived from them for association testing and will then employ these in family-based tests of association with BP disorder. He hopes to identify candidate polymorphisms, which will be a starting point for gene characterization and mutation analyses and a step towards identifying at least one causal factor in BP disorder.
